I recently bought and downloaded a locomotive pack from the download station. Does anyone know how to use it in Trainz 12? Do I have to use an existing route and substitute the locomotive?

Once the content is installed, it will act as any other locomotive in the game. You may do what you've asked, or just start a new session using the new toys.
