I agree with Phil here. "Help!" doesn't really help us when trying to help you. Remember we can't see what your problem is with Trainz, and even though you may go on to provide details in the message, your title will generally drive people away from looking at the message because it's not descriptive enough for them to come up with a quick answer, or have an idea on how to help you without opening the message.
Now back to your problem.
Open up Content Manager 3 (CM3 as we call it here).
Go to the File menu and choose Quick Database Repair.
The quick database repair isn't always "quick" so go off and do something else away from your computer while this process runs. I did this recently after importing content, and left it to run while I was a school. I came home tonight, and finished up repairing some old assets.
When this is done shutdown CM3 and reboot your computer. Try running TS010 again.
Also to ensure that the files can be written to properly, in particular if you're running under Vista or Windows 7, run the program as administrators. If you had put in this information into your post, I would simply tell you to run as an admin, but instead I had to supply extra details.
