The idea for a route can come from anywhere - a real railroad or an imaginary one, even from a video game.
A video game would be copyrighted so I would advise you to make sure that you do not use the same name for your route or make it too obvious that it is a copy. You can probably use the description "based on [name of game]" but I am not a lawyer.
