Can anyone recommend some good routes for Trainz 2010, ones that don't require any Trainz 2009 or 2006 content?

If you mean that literally (nothing but 2010 content), I don't believe there are any. If you mean routes with content that requires little or no modification to run in 2010 try East Kentucky 3, Summer-UMR 2010, Indiana Western-2010 and/or Ozark Valley Railroad. I'm sure there are others, but those are four that I'm familiar with. EK3 is available from Checkrail (http://www.checkrail.com/index.htm) and the others, if I remember correctly, are on the DLS.

You have to own T2006 and 2009 to use routes with content from them, correct?
Nope. Built-in content from previous versions is also in later versions. Owning a registered copy of a Trainz product allows you to download content for any earlier version.
